Supplements can definitely boost your metabolism. Most of them work on your basal metabolism rate and therefore you thyroid gland. You should be aware that there may be side effects to such medications and you should consult a doctor for more information about this.

It depends on the supplement.  Most supplements do not boost one's metabolism, and there is alot of marketing for diet pills that is not backed up by research.  There are only a few supplements that boost metabolism.  Some give you a slight boost (Chromium, Green Tea).  Other supplements that boost your metabolism also increase your heart rate and/or increase your body temperature (caffeine, ephedra, stimulants).  Working out boosts your metabolism more than any supplement.
